Immunosuppressive effect of Salix humboldtiana Willd. extract in response to Propionibacterium acnes

Abstract
Acne is a chronic inflammatory disease caused by blockage of the follicle due to excessive sebum production or an inflammatory response in the sebaceous glands. Propionibacterium acnes (P. acnes) is a Gram-positive anaerobic bacterium that is known to be a major factor of inflammation in acne. Salix humboldtiana Willd. (S. humboldtiana) is used for fever reducer and painkiller. It is also known to have antioxidant and anticancer activity. However, studies on the inhibitory effect of S. humboldtiana extract on the inflammatory response by P. acnes have been limited. Therefore, in this study, we investigated the possibility of anti-inflammatory effect of S. humboldtiana extract in P. acnesinduced inflammatory reaction. We found that S. humboldtiana extract successfully attenuated expression levels of pro-inflammatory mediators such as IL-1¥â, IL-6, and iNOS in P. acnes-activated mouse macrophage cell line RAW 264.7. The anti-inflammatory effect of S. humboldtiana extract was mediated by the inhibition of NF-¥êB transcription factor. Our results suggest the potential of using S. humboldtiana extract for P. acnes-induced inflammation.
